Hardy-Weinberg calculations require very large populations and is very much after the fact.
RFLP is a procedure used for determining a DNA fingerprint. A change in the fingerprint represents a change in the genome. It is very equipment intensive and has a degree of accuracy that is almost too great.
Block cells/Mitosis/Stain for banding patterns/Change in banding patterns indicates change in the chromosome content. Karyotyping is very labor and time intensive. It involves extracting individual chromosomes and the degree of accuracy is in question.
